化感物质抑制短裸甲藻的机理研究

摘要:
文章通过研究邻苯二甲酸二丁酯(DBP)对短裸甲藻的活性氧自由基(ROS,主要为H2O2)水平、过氧化标志产物丙二醛(MDA)含量以及对细胞亚显微结构的影响,初步揭示DBP抑制短裸甲藻的机理,为沿海赤潮的控制和有效防治提供有力的支持。研究结果表明,DBP通过引起短裸甲藻细胞内活性氧自由基的过度积累而引起膜质过氧化,与不添加DBP的对照组相比,在低浓度DBP(0.1 mg/L)的影响下,H2O2含量前48 h变化比较平稳,48 h以后逐渐下降。而在高浓度DBP(10 mg/L)的影响下,H2O2含量变化波动明显,暴露72 h后,H2O2含量达到248.24 nmol·(107cells)-1,约是对照组的4.67倍。过氧化标志产物MDA在低浓度DBP(0.1 mg/L)处理后暴露24 h分别出现峰值0.23 μmol·(109cells)-1、0.28 μmol·(109cells)-1,高浓度DBP(10 mg/L)处理MDA含量24 h后持续上升,72 h时高达0.351 μmol·(109 cells)-1,约是对照组的2.32倍。同时DBP可以引起短裸甲藻细胞结构受损,表现在叶绿体、线粒体等细胞器变模糊,细胞空泡化,原生质膜皱缩,细胞脂质体(黑色颗粒)明显增多等,初步认为过氧化损伤为化感物质作用机理。

Abstract:
The effect of dibutyl phthalate (DBP) on the ROS (mainly H2O2 levels),malondialdehyde (MDA) content and the degree of membrane lipid peroxidation of red tide algae Gymnodinium breve was studied in this paper,in order to reveal the material mechanism of inhibition of Gymnodinium breve and provide theoretical support,so as to effectively provide strong support for red tide control.The results showed that the H2O2 content of the exposure to 0.1 of DBP,roughly speaking,behaving steady during the beginning 48 hours and decreasing after that,while exposure to 10 mg/L of DBP fluctuating significantly which reached the peak value,248.24 nmol·(107 cells)-1,at 72 h time,3.67 times higher than the controlled.As to the MDA content,exposure to 0.1mg/L of DBP reached the peak value of 0.23 μmol·(109 cells)-1 and exposure to 1 mg/L of DBP reached the peak value of 0.28 μmol·(109 cells)-1 at 24 h time respectively.On the other hand,the MDA content of exposure to 10mg/L of DBP kept increasing after 24 h and was as high as 0.351 μmol·(109 cells)-1 at 72 h time,1.32 times higher than the controlled.By the way,ultrastructure showed that the algae cells were disorganized under DBP exposure which helped to make the primary conclusion that the damage caused by membrane lipid peroxidation was the mechanism of DBP inhibiting the growth of Gymnodinium breve.
